Minimal force is needed to aspirate and dispense the liquid, which is consistent across the full range of Pastette. Fine Tip Pastette are available in varied designs with alternative drop volumes, draw volumes and lengths to suit various tasks. Over the years, modified versions have been developed from the standard design to support customers with their application needs. These included extending the stem length to be able to reach into smaller vessels, a longer fine tip for the separation of supernatants, larger bulb volumes for filling ID strips and mini versions for small volume drop work, (which are popular) in the blood donation centres. One feature that does remain the same is the perfectly circular cross section tip, which provides precise control and accuracy of the drop volume when aspirating. The latest version, the Nanoject III, optimises sample injection efficiency to accurately deliver nanolitre volumes into both suspended and attached cells, while reducing associated cellular stress. Several features of the popular Nanoject II have been incorporated into the next generation Nanoject III. The same body design makes the Nanoject III compatible with most stereotaxic equipment. A high level of stability and precision are required in microinjection. This is crucial to minimise cellular stress and deliver the highest efficiency for improved cell/organism viability. The proven hydraulic technology of the Nanoject III ensures accurate and consistent injection, while the high torque motor significantly reduces tip movement, allowing finer manipulation. Glass micropipettes with an internal diameter of 10μm can be used with this model. The Nanoject III can be used for much lower volume injection using micropipettes with an internal diameter down to 2-3μm. Furthermore, the Nanoject III does not use any O-rings to secure the micropipette. Instead, a small chuck and silicone receiver have been incorporated, making insertion of the glass micropipette much easier. With the Nanoject III, the volume range has been expanded to 0.6nL-999.9nL, with a variable injection rate between 1nL/second200nL/second, compared to its predecessor which had a range of 2.3nL - 69nL. This capability not only extends the range of model organisms that can be investigated, but also allows investigation at multiple developmental stages using varying injection volumes. They are known to be associated with a multitude of biological functions, such as immune responses, viral pathogenicity, pregnancy, cardiovascular diseases, cell signalling, CNS-related diseases, cancer progression, and more! Exosome research has become easier with the implementation of the second generation MagCapture™ isolation kit from FujiFilm Wako, which is far superior to conventional methods, delivering higher quality, intact exosomes. The MagCapture™ utilises the phosphatidylserine (PS) affinity binding which enables the elution of intact EV’s and exosomes. Support for Exosome Research PS-Affinity Binding Method T-cell immunoglobulin domain and mucin domain-containing protein 4 (Tim4), one of the receptors for EVs binds to phosphatidylserine, which is known to be expressed on the outer surface of exosome membranes, in a calcium iondependent manner.
