The synthesis and secretion of melatonin is controlled by a circadian clock in the hypothalamus and is synchronised by the light-dark cycle. The production of melatonin is inhibited by daylight and occurs during darkness. Thus, melatonin levels are very low during day and rise in the evening. Endogenous Clock In humans, circadian melatonin profiles in body fluids (saliva, serum, plasma) provide an excellent marker for the setting of the endogenous clock. Night-time levels peak at around 3 am (acrophase) in most healthy individuals. As a general modulator of biorhythms, melatonin is involved in the timing of functions such as sleep, mood, reproduction and immune system activities. Onset of Melatonin Production The intra-individual melatonin onset and acrophase are very stable. Therefore, it is sufficient to only measure the onset of melatonin production once a whole melatonin profile has been established. The onset of melatonin production may be defined as the DLMO (Dim Light Melatonin Onset) for monitoring shifts in the profile. ELISA kits are available to measure melatonin in saliva and also its main metabolite in urine, 6-sulfatoxy-melatonin. Direct Saliva Melatonin ELISA n Sample Volume: 200μl n Sample Preparation: Pretreatment required n Time to Result: 24h n Standard Range: 0.6 - 25 pg/ml n LOQ: 1.6 pg/ml n LOD: 0.5 pg/ml Direct Saliva Melatonin Catalogue No. Description Pack Size EK-DSM Direct Saliva Melatonin ELISA 96 wells BÜHLMANN Laboratories Direct colorimetric determination of magnesium in serum, plasma and urine. Magnesium levels can be correlated to malnutrition, renal loss, malabsorption and endocrinological disturbances. Hypermagnesemia is associated with an inappropriate dosage of therapeutic agents such as magnesium sulfate, dehydration and renal disorders. Hypomagnesemia is often related to decreased calcium and potassium levels. n Linearity: up to 10 mg/dL n Open-vial stability: 90 days when stored at 2-8C Technical information: In an alkaline pH solution, magnesium ions form a stable complex with xylidyl blue, the colour intensity of which is proportional to the amount of magnesium contained in the sample. Interferences caused by calcium and proteins are minimised by a specific component and detergent. MBL is used for the determination of susceptability to infections and hence a possible requirement for the aggresive administration of antibiotics in: n Patients receiving or about to receive chemotherapy or immunosuppresive treatment n Patients with cystic fibrosis n Children with recurrent infections It may also be used as a prognastic marker for disease severity in rheumatoid arthritis and systemic lupus erythematosus. Deficiency of normally oligomerised MBL is associated with the above. Technical information: The assay is an ELISA performed in microwells coated with a monoclonal antibody against the MBL carbohydrate-binding domain. Bound MBL is detected with the same antibody that has been labelled with biotin, followed by development with horseradish peroxidase conjugated streptavidin with a chromogenic substrate. The assay is a four-step procedure.
